信号通路(Signal pathway)是指能将细胞外的分子信号经细胞膜传入细胞内发挥效应的一系列酶促反应通路。一个信号通路的构成主要包括三部分元件,即受体(receptor)与配体(ligand)、蛋白激酶(kinase)、转录因子。在信号传导过程中,配体与细胞膜表面或细胞内受体结合,蛋白激酶通过磷酸化改变下游蛋白构象从而对一系列下游蛋白进行调节并将信号级联放大,最终通过转录因子对基因转录进行调节最终产生一系列细胞的综合应答。 信号传导过程是细胞对外界刺激产生反应并最终引发特异性生物学效应的有效方式。几十年的基础研究逐渐解释了许多在癌症、心血管疾病、自身免疫性疾病等疾病发生与发展过程中起关键作用的信号通路。同时,针对信号通路的药物筛选与开发也成为了疾病免疫治疗的新思路。
JAK-STAT信号传送途径是一个进化保守的信号级联反应,介导对细胞因子和生长因子的反应,使得细胞外的化学信号跨越细胞膜并将信息传送到细胞核内DNA上的基因启动子,最终引起细胞中DNA转录与活性水平发生改变,是除了第二信使系统外最重要的信号途径。JAK-STAT系统由三个主要部分组成:受体、JAK激酶、信号转导及转录激活蛋白(STAT)。
